Purification and characterization of nitrile hydratase from Rhodococcus rhodochrous M8

摘要
The nitrile hydratase of Rhodococcus rhodochrous M8 has been purified and the enzyme properties studied. The molecular mass of the enzyme was 510 kDa. The nitrile hydratase was composed of two different types of subunits with molecular masses 23.5 kDa and 28 kDa. Optimal temperature and pH values for the enzyme activity and stability were: 20-30 degrees C, pH 7.0-7.2. The nitrile hydratase activity was stabilized by butanoic acid. The enzyme exhibited high activity towards aliphatic substrates. The aminoterminal amino acid sequence was obtained.
