PI/DAPI Fluorochrome Staining Technique for Identifying Citrus Chromosomes

摘要
The mitotic metaphase chromosomes of several citrus were stained with propidium iodide (PI) and (DAPI). The stable chromosome patterns were observed with the suitable proportion of PI/DAPI volume. Based on the relative-sized PI signals and the morphology of no-signal chromosomes, 18 chromosomes of citrus were classified into 7 groups. In addition, the relative-sized PI signals on the chromosomes were observed maintain stable in the cell cycle. It is considered that the chromosome combined with PI fluorochrome were highly stable construction.
