Inhibition of 2-oxoglutarate dehydrogenase complex by free fatty acids

Fatty acids, along with carbohydrates, are the main sources of fuel for the Krebs cycle in the form of acetyl-CoA. They have a regulatory role in liver and heart. We studied the possibility of regulation of the activity of 2-oxoglutarate dehydrogenase complex (OGDC) and its first component, oxoglutarate dehydrogenase (OGD), in bovine heart by free long chain fatty acids. The results indicate that oleate plays a significant role in OGDC and OGD function by changing kinetic parameters of the complex. This fatty acid inhibits oxoglutarate dehydrogenase complex activity with 2-oxoglutarate by increasing [S](0.5) values.
