Rho GTPases regulate degranulation and cytokine release from RBL-2H3 cells

摘要
Clostridium difficile Toxin B (toxin B), which inactivates Rho GTPases, Rho, Rac, and Cdc42, inhibited the release of TNF-alpha as well as degranulation from antigen-stimulated RBL-2H3 cells, independently of reorganization of actin microfilaments. Rho-associated kinase (ROCK), one of the downstream effectors of Rho, was selectively involved in the release of TNF-alpha but not in degranulation. Toxin B inhibited the translocation of Ca2+-dependent protein kinase C (alpha and beta isozymes), which plays an important role in the release of TNF-alpha as well as degranulation. These results indicated that Rho GTPases regulate the release of TNF-alpha and degranulation by regulating at least the activation of Ca2+-dependent PKC in antigen-stimulated RBL-2H3 cells.
