Preparation and in vitro release of magnetic citosan-5-fluorouracil nanoparticles

In order to prepare the targeting drug-loaded system, magnetic chitosan-5-Fu nanoparticles (MCN-Fu) were prepared by using 5-Fu as a model drug, Fe3O4 nanoparticles as the core and glutaraldehyde as cross-linking agent under ultrasonic treatment. TEM images show that MCN-Fu are spherical shape with diameters ranged from 50 nm-60 nm. The IR results indicate there is a strong molecular interaction between 5-Fu and chitosan. The drug content, in vitro release and magnetic properties of MCN-Fu were investigated by UV-vis spectrum. The results indicate that the drug content is 21.3%, and accumulative release percentage in phosphate buffer (pH = 7.2) solution is 67.6% in 30 h, the drug release rate is found decreased and they can in some degree reduce initial burst. MCN-Fu have good magnetic properties and target effect.
