A method for isolating DNA-specific catalytic antibodies (natural abzymes) is described Purification procedures include affinity chromatography on protein A Sepharose or DNA-cellulose (the latter using either native DNA or glutaraldehyde-modified DNA as sorbent) and gel-permeation HPLC. Elution conditions were optimized to ensure maximum yields of activity. Biochemical and immunochemical characteristics of the natural DNA-specific abzymes are reported Their interaction with a model substrate and the influence of different effectors on DNA hydrolysis were investigated.
