摘要
DNA damage and repair measurement by alkaline sucrose gradient sedimentation was adapted to in vivo biopsy samples by refining a sensitive DNA fluorescence assay and establishing quantitatable sedimentation conditions for high-MW DNA from isolated nuclei. Induction and repair of X-ray-induced single-strand breaks plus alkali-labile bonds in rat liver DNA were measured in whole liver and in successive biopsies taken from single animals; repair half-time was 20-30 min. Incubation of UV-irradiated rat liver nuclei with UV endonuclease before sedimentation enabled detection of pyrimidine dimers.
