摘要
Ziziphus jujuba Mill. Cv. Zanhuangdazao Hod. is a triploid cultivar in natural. There are two ploidy level in its pollens, haploid and diploid. To obtain the regenerated plantlets from pollens, we studied the effects of basal medium, type of carbon source, darkness and cytokinin in anther culture of Ziziphus jujuba Mill. Cv. Zanhuangdazao Hod.. The callus induction and shoot regeneration of anthers were obtained on the same medium, i.e. MS (Murashige and Skoog) (1962) medium supplemented with maltose 20g/L 2,4-dichlorophenoxyacetic acid (2,4-D) 1.0 mg/L and silver nitrate (AgNO3) 1.0 mg/L in the dark for 30 days with the highest percentage of anther explants that produced calli (79.3%), overall efficiency of regeneration (72.6%). By flow cytometry analysis of ploidy level, we first obtained the diploid shoots from pollens of Ziziphus jujuba Mill. Cv. Zanhuangdazao Hod, successfully.
