A new host cell-line for the fast, reproducible and efficient production of biopharmaceuticals

摘要
We describe here the proof of concept of a new, innovative "wildcard"-strategy (termed TaCEx-system, tag, characterize and exchange) for the generation of CHO-based cell-lines for the production of biopharmaceuticals using site-specific recombinase-mediated cassette exchange. Core element of the TaCEx-system is a pre-formed host cell-line. It contains a tagged reporter cassette by which the expression capacity of the integration site has been characterized and which allows the exchange of the reporter against a gene of interest (GOI). Such a new host cell facilitates the cloning of a GOI into predetermined genomic expression hot spots within days in order to get reproducible, highly efficient production cell-lines.
