Molecular Cloning and Expression of a Pathogenesis-Related Protein Gene in Citrus Fruit of Peel Pitting

摘要
In a search for differentially expressed genes in peel pitting of citrus fruit, a cDNA subtraction library was constructed and a sequence encoding pathogenesis-related (PR) protein was isolated. Based on sequence information derived from this fragment, a full-length cDNA (CsSPR, GenBank accession no. EF010853) of 679 nucleotides was isolated from citrus fruit. This cDNA contains a 483-bp open reading frame encoding a protein of 161 amino acids with a calculated molecular mass of 17.60 kDa and an isoelectric point was 5.74. The deduced protein contained the conserved Bet v I domain of PR proteins. Expressions of CsSPR gene in healthy and pitted citrus fruits were checked and results showed that CsSPR mRNA levels in pitted fruits were higher than those in healthy fruits. The present results suggested that CsSPR is a peel pitting related gene of citrus fruit.
