Isolation of genomic DNA from wild species of Kuwaiti flora using different extraction methods

摘要
Fifty one wild species of Kuwaiti flora were subjected to six different standard protocols for DNA extraction, with the view of selecting the one yielding the most suitable and consistent method for further molecular studies. The comparison between the methods for the DNA analysis were based on the purity, yield, response to restriction enzymes, response to RAPD-PCR and efficiency in the removal of some secondary products. The method of Dellaporta et al. (1983) was more effective than the others in terms of showing a high number of DNA extracts that have relatively high levels of purity, yield, PCR-products, enzyme digestion and absence of secondary metabolites. On the other hand, the method by Rogers & Bendich (1985) gave DNA extracts from almost all species used that showed low levels of purity, yield, PCR-products and enzyme digestion. This latter method however was effective in elimination of secondary metabolites from a large number of species tested.
