CONSTRUCTION OF A RECOMBINANT LENTIVIRAL VECTOR CONTAINING MIR-210 AND ITS STABLE EXPRESSION IN HMEC

摘要
Recent research indicates that miR - 210 is involved in various biological processes of blood vessel endothelial cell. In present study, we report the construction of recombinant lentivirus miR210 expression vector and its stable expression of miR - 210 in transfected human microvascular endothelial cells (hMECs). The precursor of human miR - 210 gene was amplified by PCR and the PCR products were linked with eukaryotic expression vector pGCSIL-GFP. Through packaging by 293T cell, the recombinant lentiviral expression vector of miR - 210 and its controls were transfected. to hMECs. Fluorescence detection showed that fluorescence intensity of GFP was highest between 48 and 72 hours post-infection, and qRT-PCR showed an increase in miR - 210 expression in recombinant lentivirus transfected cells compared with the expression in its controls. The recombinant lentiviral expression vector of miR - 210 was constructed successfully, which may facilitate further study of blood vessel endothelial cell.
