Anticonvulsant activities of antagonists of NMDA and calcium-permeable AMPA receptors in a model of maximum electroshock in rats

摘要
It is known that activation of ionotropic glutamate receptors plays a leading role in the initiation of seizures and their spreading; however, contribution of one of the subtypes of these receptors, calcium-permeable AMPA receptors (Ca-AMPAR) to seizures is practically unknown. The anticonvulsant action of two Ca-AMPAR blockers, IEM-1754 and IEM-1925, as well as the combination of IEM-1925 with the NMDA channel blocker IEM-1921, was studied in Wistar rats using the maximum electroshock model. It was shown that two Ca-AMPAR blockers are unable to prevent the development of generalized tonic seizures that were induced by electroshock. The combination of IEM-1921 and IEM-1925 did not have a stronger anticonvulsant action than IEM-1921 alone. These results reflect the weak involvement of Ca-AMPAR in the realization of tonic seizures induced by the maximum electroshock.
