Genetic diversity of Pleuronectes yokohamae population revealed by fluorescence microsatellite labeled

摘要
The genetic diversity of two Pleuronectes yokohamae population was studied by 17 fluorescently labeled (FAM, HEX, or TAM) microsatellite primers. One wild population was from North Korea with a sample size of 5 individuals. The other cultured population was from a fish farm in Yantai China with a sample size of 20 individuals. Totally 221 alleles were obtained from the two population. Average PIC (polymorphic information content) was (>0.5) that means all the loci were higher polymorphic and could be applied to genetic structure analysis effectively. The average allele numbers in wild and cultured population were 5 and 8 respectively. The mean H-e of two populations were 0.8118 and 0.77 while the mean H-0 were 0.6824 and 0.6263. The PIC value ranged from 0.530 to 0.8230 in the wild population and 0.375 to 0.8392 in the cultured populations. Only one loci (PTH8) in cultured population was under the HEW and all the other locus were deviated from HEW due to heterozygosity excess. The mean Nei's Fst value, genetic identity and genetic distance (D) between two populations were 0.1276, 0.1637 and 1.8095 respectively. Under S.M.M. model, cultured population had appeared bottleneck (P < 0.05), but under I.A.M. model, no populations have appeared bottleneck. All the results showed that genetic diversity of wild and cultured population was almost at same level and there existed genetic differentiation between two populations. (C) 2014 Elsevier Ltd.
