Formation of DNA double-strand breaks and DNA-protein crosslinks by irradiation of DNA in the presence of a protein

摘要：
Double-stranded DNA and serum albumin were irradiated with X-rays in different phosphate solutions under N-2, N2O and air. Double-strand breaks (dsb) were determined by electrophoresis. A new stochastic model makes now possible to calculate the values of DNA-protein crosslinks (Dpc) with improved accuracy from the nitrocellulose filter assay. The yields of Dpc are in the same order of magnitude as the yields of dsb. The conclusion is that secondary protein radicals and protein peroxyl radicals cause DNA dsb and Dpc.
