摘要
An HPLC-MS/MS analytical method for quantitative determination of acetylsalicylic acid (ASA) as an antithrombotic agent and its metabolite salicylic acid (SA) in human blood plasma was developed and validated. The optimum concentration ranges were 0.020 - 1.200 mu g/mL for ASA and 0.200 - 12.00 mu g/mL for SA. The optimum conditions for sample preparation using liquid-liquid extraction by EtOAc followed by removal of the solvent in a stream of N-2, redissolution of the dry residue (to concentrate the sample and increase the method sensitivity), chromatography, and mass-spectrometric detection were selected. The developed method could be used further for pharmacokinetic and bioequivalence studies.
