A novel preparation and X-ray crystallographic analysis of phospha sugar nucleoside derivatives

2-Azido-3-hydroxy-3-methyl-1-phenylphospholane 1-oxide was obtained by the substitution reaction of the corresponding 2-bromo-3-hydroxy-3-methyl-1-phenylphospholane 1-oxide, which was derived from 3-methyl-1-phenyl-2-phospholene 1-oxide being an unsaturated heterocyclic phosphorus compound. Reaction of the azido-phospholane derivative with some alkynes afforded novel 3-hydroxy-3-methyl-2-triazolyl-1-phenylphospholane 1-oxide derivatives whose structures were analogs of nucleosides of phospha sugar derivatives. The structure of the triazole derivatives thus prepared was analyzed by X-ray crystallography. Hydrogenolysis of the 2-azido-1-phenylphospholane 1-oxide derivative with Pd/C under atmospheric hydrogen afforded 2-amino-3-hydroxy-3-methyl-1-phenylphospholane 1-oxide. The 2-amino- and 2-azido-phospholane derivatives prepared were analogs of N-glycosides of phospha sugar derivatives.
