Colonic bacteria regulate epithelial expression of the polymeric immunoglobulin receptor through MyD88-dependent signaling

The polymeric immunoglobulin receptor (pIgR) promotes intestinal homeostasis via epithelial transcytosis of protective IgA antibodies. We hypothesized that colonic bacteria modulate pIgR expression via MyD88-dependent signaling. Co-culture of HT-29 human colonic epithelial cells with live or heat-killed E. coli induced comparable expression of pIgR mRNA. By contrast, live E. coli induced higher expression of the pro-inflammatory factors IL-8 and TNF. Live and heat-killed E. coli also induced expression of the negative regulatory factor A20. Levels of pIgR and mucosal IgA were significantly reduced in colons of mice deficient in the signaling adaptor MyD88. Our results indicate that interactions of commensal bacteria with epithelial cells regulate pIgR expression.
