Using a Commercial Method for Rubisco Immunodepletion in Analysis of Plant Proteom

Rubisco immunodepletion was tested using a Seppro IgY-Rubisco spin column in 2D-PAGE-based proteomic analysis. The column yield was analyzed in 20 consecutive runs and the depletion from tissues with higher and lower Rubisco contents was evaluated. An increase in the number of detected protein spots of up to 25 % was found. This procedure allows to follow the profiling of a similar number of protein spots in Rubisco-depleted proteom resolved on small gels as is usual with standard total protein extracts resolved on large gels. We found this procedure especially suitable for experiments with a limited amount of the starting material.
