Optimizing effective factors on alkaline protease production by Bacillus lichiniformis under submerged fermentation

摘要
Aim. Alkaline proteases play an important role in industrial processes and produced by a bacillus lichiniformis. Methods. In this study, the suitable composition of bacterial growth and optimum conditions of protease production were determined. Results. Maximum activity(34130 U/mL(-1))was obtained when the cell were grown under the submerged fermentation at in a medium containing: mixture of soybean meal and casein (20 gl(-1)), mixture of molasses and corn starch (50 g/L-1), K2HPO4, 4; Na2HPO4, 1; CaCl2, 0.05; Na2CO3, 8 at 38 degrees C for 48 hour incubation period with agitation of 300 rpm. The optimum pH and temperature of the enzyme were around 7.5 and 38 degrees C respectively. It was found that mixture of soybean meal and casein have suitable conditions for protease production. Conclusion. On the basis of the results, the best condition of protease production was as follows: pH=7.5, temperature=38 degrees C, rotation of agitator=300 rpm. It can be concluded that this enzyme has wide application in various industrial and medical fields and it can be produced in large scale from the bacillus lichiniformis with apply cheap and easily available medium substrate by fermentation technology
