A study of the expression and intracellular localization of the protein TspO in the phototrophic bacterium Rhodobacter sphaeroides

摘要
The expression of the gene tspO, which encodes the 17-kDa protein TspO, has been studied. TspO is unusually rich in aromatic amino acids, L-tryptophan in particular. TspO of the phototrophic bacterium Rhodobacter sphaeroides is homologous to the mitochondrial benzodiazepine receptor of animals. TspO was expressed in E. coli cells, purified, and used as an antigen for obtaining polyclonal antibodies. The antibodies specifically reacted with TspO of the outer membrane fraction of R, sphaeroides. The transition of the cells from aerobic to anaerobic (photosynthetic) type of metabolism increased the level of TspO severalfold. Possible mechanisms of regulation of the physiological activity of TspO in R. sphaeroides are discussed.
