Detection of biomarkers for liver fibrosis using high-throughput electrochemical microimmunosensor

摘要
A high-throughput electrochemical microimmunosensor for the detection of biomarkers for liver fibrosis was developed. The antibodies, hyaluronic acid binding protein (HABP), lamin antibody (a-LN) and type IV-collagen antibody (a-IVC). are immobilized on different electrodes of the microelectrode array by copolymerizing into the partly insulated poly(o-phenylenediamine) by means of cyclic voltammetry. Electrochemical detection of the corresponding antigen was based on the extent of electrode Insulation toward a redox probe (ferrocenemethanol) solubilized in the electrolyte as a result of the formation of the antigen-antibody complex at the electrode surface. The microimmunosensor exhibits enough sensitivity to detect the three biomarkers at a concentration level down to 3 ng/mL. The microimmunosensor has been applied to real samples, the results agree well with those obtained by radioimmunoassay (RIA). With the possibility of being portable and considering its ease of use, robustness., and simplicity. the microimmunosensor has great potential as a tool for the screening and early detection of liver fibrosis.
