Construction of a Molecular Genetic Linkage Map for Longan Based on RAPD, ISSR, SRAP and AFLP Markers

摘要
A large F-1 hybrid population was created by crossing a high quality cultivar 'Fengliduo' with a large fruit size main cultivar 'Dawuyuan'. Among the hybrid F-1 population (375 individuals), 94 individuals were randomly chosen and used as the mapping population. Based on the mapping population, a molecular genetic linkage map of longan was constructed with RAPD, ISSR, SRAP and AFLP molecular markers. According to the linkage analysis with JoinMap3.0, the molecular linkage map of 'Fengliduo' consisted of 21 linkage groups, which involved 183 markers with an average interval of 5.84 cM and covered a total distance of 965.1 cM; while the molecular linkage map of 'Dawuyuan' fell into 22 linkage groups, which involved 251 markers with an average interval of 4.65 cM and covered a total distance of 1064.8 cM of longan genome. This was the first report about high-density molecular genetic map for longan and would lay a solid foundation for gene location and map assisted selection in longan breeding.
