Pancreatic ductal glandular carcinoma patients often feature with a poor prognosis, especially, the five-year survival rate, including all diagnostic stages combination, is only less than 5%. Even though advances in surgery have significantly augmented the survival rates, the clinical outcomes remain relatively low. Chemodynamic therapy and photothermal therapy exhibit an effective and relatively high safety tumor elimination modality. Here, we have engineered Fe3O4 nanocrystals with Au nanozyme and tetra-sulfide bonds incorporated in mesoporous silica shell coating, then after surface functionalization of RGD, a sequentially GSH triggered nanoplatforms is obtained with accurate tumor recognizing and intelligent Au nanozyme release profile. It is capable of mediating glycolysis reaction which is widely considered as a promising strategy to self-supply the intracellular H2O2 in situ. The bare Fe3O4 nanocrystals can certainly destroy the tumor cells through photothermal therapy and under near-infrared laser illumination (808-nm laser). Collectively, both hyperthermia and generated H2O2 could significantly improve the Fenton reaction of Fe3O4 nanoparticles so that an excellent antitumor effect can be achieved both in vitro and in vivo. Combined with the superior biosafety feature, this work provides a practical technique for enhancing drug delivery efficacy and malignant tumor elimination.
