Development of an efficient in vitro regeneration system in broccoli (Brassica oleracea L. var. italica), a highly recommended vegetable crop

In the present study, different explants of broccoli (Brassica oleracea L. var. italica) were tested for their ability to regenerate in vitro shoots. Hypocotyl, cotyledon, leaf and petiole explants of seven days old seedlings were tested for shoot regeneration on medium containing different concentration and combinations of BAP and NAA. Highest regeneration frequency was achieved by petiole explants (90.33%) followed by cotyledon (89.78%), hypocotyl (88.78%) and leaf (64.51%) on MS medium supplemented with 4.5 mg/l BAP + 0.5 mg/l NAA, 3 mg/l BAP + 0.5 mg/l NAA, 3.5 mg/l BAP + 0.25 mg/l NAA and 4 mg/l BAP + 0.5 mg/l NAA, respectively. The highest rooting frequency (99.81%) with more number of adventitious roots were obtained on half strength MS medium containing 0.1 mg/l IBA. The well-developed rooted plantlets were successfully transplanted into pots containing cocopeat: perlite: vermiculite (3:1:1) mixture and allowing to grown under greenhouse conditions. Therefore, in the current study, an efficient protocol has been standardized, which can be efficiently used further for genetic transformation experiments in broccoli and other Brassica spp. to develop abiotic and biotic stress tolerance in plants.
