胶质母细胞瘤病人SOX21基因与预后的关系

摘要
目的 通过对癌症基因组图谱数据库的生物信息学分析,探讨SOX21基因的差异表达在胶质母细胞瘤病人中的预后价值及临床意义。方法 采用Wilcoxon非参数检验比较SOX21基因在胶质母细胞瘤组织和非肿瘤组织中的表达,采用Logistic回归分析SOX21与临床病理特征的关系。进一步采用Kaplan-Meier和Cox回归分析SOX21基因与胶质母细胞瘤病人生存概率的相关性。最后,通过基因集富集分析对SOX21基因的生物学功能进行注释。结果 SOX21基因在胶质母细胞瘤中的表达与亚型显著相关,与经典型相比,间质型、神经元型和前神经元型的差异有统计学意义(P<0.05)。Kaplan-Meier生存分析显示,SOX21基因表达下调的胶质母细胞瘤病人预后较SOX21基因表达上调的胶质瘤差(P=0.003)。单因素分析表明,SOX21基因表达上调与良好的总生存(OS)相关[HR=0.69,95%CI(0.54,0.88),P=0.003];多因素分析显示,SOX21基因也与胶质母细胞瘤的OS独立相关[HR=0.70,95%CI(0.55,0.90),P=0.005]。基因集富集分析显示,丙酸代谢、缬氨酸、亮氨酸和异亮氨酸降解、过氧化物酶体和赖氨酸降解均集中于SOX21高表达表型。结论 SOX21表达上调可能是胶质母细胞瘤病人良好生存的潜在预后生物标志物。此外,丙酸代谢、缬氨酸、亮氨酸和异亮氨酸的降解、过氧化物酶体和赖氨酸的降解可能是SOX21基因在胶质母细胞瘤中调控的关键信号通路。
