Antioxidant activity of superoxide dismutase from wheat leaves

The antioxidant activity of superoxide dismutase (SOD) from wheat leaves has been verified within the animals in vivo as well as in vitro methods. DPPH and hydroxyl free radical scavenging capacity and the reduction capability of SOD solution from young wheat leaves have been determined by means of DPPH·assay, Fenton reaction and Fe3+ reduction method with in vitro experiment respectively. The vivo experiment was promoted as following: a total of fifty healthy Kunming mice were randomly divided according the feeding solvent different into dose group and control group. The dose group and control group were intragastrically administered with SOD tested samples and equivalent solvent respectively. The capability of hydrogen peroxide enzyme (CAT), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) content in liver tissue were measured. The results indicated that SOD enzyme fluid had good antioxidant removal ability through in vitro experiments, scavenging capacity could reach 96.95% and 98% in SOD (1 mg/mL) enzyme fluid at 1 mL level of DPPH· and hydroxyl free radical respectively. Reducing capacity showed an increasing trend. Compared SOD dose group with the control group in vivo tests, the significant difference (P<0.05) MDA concentration could be determined in mice liver tissue, and the content of every dose group formanced reducing trend. Higher SOD dose could improve GSH-Px vitality and CAT (P<0.05) activity in mice liver tissue. Therefore, obvious antioxidation activity was observed on superoxide dismutase from younger wheat leaves through in vitro and in vivo methods.
