Two-step pretreatment induces organogenesis from recalcitrant blueberry cultivar

It has been found that leaf explants from 2-week-old shoot cultures of 'Bluecrop' can be induced to regenerate shoots by using a two-step pretreatment. The protocol involves culturing the explants on a medium containing 5 μM thidiazuron and 2.6 μM NAA, transfer to a medium containing 7 μM zeatin riboside and 2.6 μM NAA, transfer to regeneration medium containing 1 μM thidiazuron, and transfer to medium without growth regulators for 10 days. In contrast, no shoot regeneration occurs if shoots are cultured on regeneration medium without pretreatment.
