Solid-phase extraction coupled with LC-MS for the determination of fentanyl analogs in biological samples

A solid-phase extraction (SPE) coupled with liquid chromatography-mass spectrometry (LC-MS) method was established for the determination of 10 fentanyl analogs in serum and urine samples. The sample solution was extracted by the C18 SPE column. Then, the analytes on SPE column were eluted with methanol. The eluate was collected and determined by LC-MS. For the urine matrix, the linearity range of each fentanyl analog was 1-100 μg / L with R2 higher than 0. 99. The limits of detection (LODs) and limits of quantification (LOQs) were 0. 18-0. 32 μg / L and 0. 61-1. 00 μg / L, respectively. For the serum matrix, the linearity range of each fentanyl analog was 2-100 μg / L with R2 higher than 0. 99. The LODs and LOQs were 0. 15-0. 33 μg / L and 0. 50-1. 00 μg / L, respectively. The extraction recovery rates in urine and serum were 84. 2%-107. 7% and 84. 7%-99. 7%, respectively. No significant matrix effect was observed. This method can satisfy the requirement for simultaneous analysis of various fentanyl analogs in biological samples. © 2021, Youke Publishing Co.,Ltd. All rights reserved.
