Production and Optimization of Xylanase by Thermophilic Bacillus sp Isolated from Soil

Totally 15 strains of thermophilic Bacillus spp. were isolated from soil and five of the thermophillic bacteria showed xylanolytic activity on secondary screening based on the diameter of the clear zone formation on congo red xylan. agar. On the basis of morphological, cultural, biochemical and by molecular characteristics the potential xylanolytic Bacillus sp. was identified as Bacillus pumilus. For maximum xylanase production B. pumilus required an optimum temperature of 50 degrees C for 72 h. The optimum pH observed was 7.0 +/- 0.2. Xylan concentration at 1.0% in the production medium was found to be optimum. Production media with 1% galactose as carbon source, 1% beef extract as nitrogen source and 0.5% FeCl3 as mineral salt enhances the xylanase production.
