STRUCTURAL AND FUNCTIONAL STATUS OF ERBB-2, HRAS-1, C-MYC ONCOGENES, P53, RB1 ANTIONCOGENES, AND DELETIONS OF CHROMOSOME-17 LOCI IN BREAST CARCINOMAS

Molecular-genetic abnormalities were found in 27 surgical specimens of 44 ones obtained from patients with the breast carcinoma. Amplification of oncogene ERBB-2 was observed in 13 (30 %) cases, c-myc - only in one tumor. Loss of heterozygosity of HRAS-1 oncogene was detected in 4 of 21 specimens. Deletions of anonymous probes YNZ-22 (17 p) were found in 14 of 29 heterozygous specimens. THH-59 (17 q) - in 7 of 26. Rearrangement of p53 was detected in one specimen. There were no abnormalities in the structure of oncogene ERBB-1 and antioncogene Rb1. An analysis of RNA in 32 neoplasms has revealed more than 5-fold overexpression of ERBB-2 in 9 specimens and RB1 - in 2 ones, but absence of high superexpression of oncogenes ERBB-1, HRAS-1, jun and antioncogene p53. Amplification of FRBB-2 was associated with 17 p deletions. Surprisingly, 17 p deletions also strongly correlated with the absence of nodal involvement, that might be caused by the losses of metastasis-associated genes.
