FLUOROMETRICAL METHOD OF COMBINED DETERMINATION OF CHLOROPHYLL-A, CHLOROPHYLL-B AND PHAEOPHYTIN-A

Method of fluorometrical determination of chlorophylls << a >> and <<b>> in mixture by means of two interference filters in described. The method was worked through on chromatographically pure pigments. Equations for determination of separate green pigments concentration were calculated by means of stable calibrating coefficients were derived for our fluorometer. Simple method of registration of chlorophyll <<b>> influence on the phaeophytin <<a>> determination by means of fluorescent method. It is important, as when the correlation of chlorophylls <<b>>/<<a>> = 0,4, determination of artefact increases up to 30%. The method is recommended as express - analysis of the determination of the native chlorophyll and phaeophytin concentration in natural photoplankton populations.
