基于NF-κB信号通路探讨枸杞多糖对过敏性鼻炎小鼠Th1/Th2细胞因子的影响

摘要
目的 探讨枸杞多糖调控核因子-κB(NF-κB)信号通路对过敏性鼻炎小鼠Th1/Th2细胞因子的影响。方法 将72只SPF级BALB/c小鼠随机分为为正常组、模型组、丙酸氟替卡松组及枸杞多糖低、中、高剂量组,每组12只。除正常组外,其余各组小鼠均采用卵清蛋白致敏法构建过敏性鼻炎模型。建模成功后,丙酸氟替卡松组每天予以丙酸氟替卡松滴鼻,枸杞多糖低、中、高剂量组每天予以25 mg/kg、50 mg/kg、100 mg/kg的枸杞多糖灌胃,正常组与模型组予以等量生理盐水灌胃,均持续干预21 d。比较各组小鼠鼻部症状评分,HE染色观察鼻黏膜组织病理学形态,ELISA法检测血清干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)、免疫球蛋白E(IgE)水平,Western blot法检测鼻黏膜组织中Toll样受体4(TLR4)、NF-κB p65和磷酸化NF-κB p65(p-NF-κB p65)蛋白表达情况。结果 与正常组比较,模型组小鼠鼻部症状评分明显升高(P<0.05);鼻黏膜组织中有较多嗜酸性粒细胞浸润,腺体增生;血清IL-4、IgE水平及鼻黏膜组织中TLR4、p-NF-κB p65蛋白相对表达量均明显升高(P均<0.05),血清IFN-γ水平明显降低(P<0.05)。与模型组比较,各给药组小鼠鼻部症状评分明显降低(P均<0.05);鼻黏膜组织炎性反应和腺体增生均明显减轻;血清IL-4、IgE水平及鼻黏膜组织中TLR4、p-NF-κB p65蛋白相对表达量均明显降低(P均<0.05),血清IFN-γ水平均明显升高(P均<0.05)。且随枸杞多糖剂量的增加,各指标改善越明显(P均<0.05)。结论 枸杞多糖可以通过抑制TLR4/NF-κB信号通路相关蛋白的表达,纠正Th1/Th2细胞因子失衡,减少IgE的生成,进而有效减轻过敏性鼻炎小鼠的鼻黏膜炎症反应。
