雷公藤红素靶向HSDL2蛋白促进乳腺癌MDA-MB-231细胞凋亡(英文)

乳腺癌仍然是目前威胁女性健康的最常见的癌症类型之一。其中,三阴性乳腺癌(TNBC)是指一种异质性乳腺肿瘤,即缺乏人类表皮生长因子受体2(HER2)、孕激素受体受体(PR)和雌激素受体(ER),目前仍无最佳治疗方法和治疗靶点。因此,为TNBC寻找新的治疗靶点和筛选基于新靶标的候选药物变得迫在眉睫。羟基类固醇脱氢酶2(HSDL2)属于短链脱氢酶/还原酶(SDR)超家族,位于9q32染色体位点,在多种类型的肿瘤中异常表达,但其在乳腺癌治疗中的作用机制尚不清楚。雷公藤红素是从传统中草药雷公藤中提取的一种五环三萜化合物,在乳腺癌细胞中抗肿瘤活性的分子机制尚不完全清楚。为合理开发利用雷公藤红素抗乳腺癌提供充分科学依据,本研究采用基于活性的化学蛋白质组学方法研究雷公藤红素在乳腺癌细胞中的蛋白靶点和分子机制。研究结果表明,低浓度雷公藤红素具有抗肿瘤作用,可直接与HSDL2结合并抑制其表达。促凋亡蛋白Ba X的表达增加,而抗凋亡蛋白Bcl-2的表达水平降低,凋亡率升高。用si-HSDL2转染细胞后,细胞凋亡率与给药后相似。然而,过表达HSDL2后,细胞凋亡被逆转。总之,我们的研究结果表明雷公藤红素可能通过靶向HSDL2从而促进细胞凋亡。此外,质谱数据表明HSDL2与MAPK信号通路之间存在关系,HSDL2的表达与ERK的磷酸化程度直接相关。
