A New Piperidine Derivative with Antiserotonin Anti-Migraine Activity and Its Combination with Picamilon

A new serotonin antagonist with anti-migraine activity, 2,3,4-trimethoxy-N-(2,2,6,6-tetramethylpiperidin-4-ylidene)benzohydrazide hydrochloride (LK-1033), was identified as a result of screening piperidine derivatives. Experiments on non-linear rats with registration of local blood flow in the cerebral cortex and blood flow in the carotid artery revealed the ability of LK-1033 to attenuate changes in cerebral blood flow caused by serotonin. However, this compound lowered the initial level of cerebral circulation. Combination of LK-1033 with the anti-ischemic drug picamilon was proposed to neutralize this negative effect. The combination containing LK-1033 and picamilon exhibited both antiserotonin cerebrovascular activity and antiischemic activity.
