Establishing Method and Evaluation Indexes of λ-Carrageenan Induced Acute Inflammatory Paw Edema Model in C57BL/6J Mice

Inflammatory response is a complex process of the body as it delivers the appropriate defense against harmful stimuli. lambda-carrageenan induced paw edema mice model is also known to be used in study for anti-inflammatory activity. Therefore, we established lambda-carrageenan induced acute paw edema in C57BL/6J mice and detected a variety of indexes for more roundly evaluating the effect of anti-inflammatory drug. Mice were stimulated with lambda-carrageenan for the induction of inflammatory model and orally treated with indomethacin (10 mg/kg), which as a positive drug, should be injected 1 h before lambda-carrageenan injection. Mice were divided into three groups: control group, lambda-carrageenan (CAR) group and lambda-carrageenan + indomethacin (CAR + INDO) group. Evaluation indexes of the inflammatory response in mice model were detected by paw thickness, histological analysis, serum levels of cytokines in vivo. Results showed that paw thickness was seriously swelling in CAR group, moreover it was significantly improved in CAR + INDO group compared with that in CAR group. Histological analysis also indicated that the obvious inflammatory cell infiltration and synovial hyperplasia were found in the CAR group, whereas the CAR + INDO group revealed mild clinical symptom expression. The concentration of tumornecrosis fatoralpha (TNF-alpha), interleukin-1 beta (IL-1 beta) and interleukin-17 (IL-17) in CAR + INDO group were significantly reduced compared with those in CAR group. We established acute inflammatory model and roundly evaluated a variety of indexes in C57BL/6J mice, which exhibited excellent stability and repeatability. Our research has provided a systematic research method for the study of screening and evaluation of activity in anti-inflammatory drug.
