Online Determination of Oxygen Uptake and Carbon Dioxide Production Rates in Mammalian Cell Culture Using Mass Spectrometry

摘要
In this investigation oxygen uptake rates (OUR) of three fed-batch processes are studied in large scale (2000 L, process A and B) and in small scale (40 L, process C) using on-line off-gas quadrupole mass spectrometry. The OUR profiles are in good agreement with typical culture parameters such as viable cell concentration, product yields, and the stoichiometric ratio. YO2/Glucose of oxygen uptake to glucose consumption. Theoretically 1 mol glucose requires 6 mol oxygen for complete oxidation under ideal culture conditions. Then glucose is effectively utilized and build-up of metabolic by-products such as lactate is minimized. By this means increased monoclonal antibody (mAb) product yields are possible.
