Study on the behavior in capillary electrophoresis of the programmed cell death process in Saccharomyces cerevisiae

摘要
In this paper, the mode of the programmed cell death (PCD) in yeast Saccharomyces cerevisiae induced by acetic acid was investigated with the flow cytometric (FCM) and scanning electron microscopy (SEM). It was, found that after 21 h of treatment with 800 mmol/L acetic acid, there was a hypodiploid peak of the induced yeast cell in FCM,and the cell surface was wrinkly and sunken in SEM, respectively. In the studying on the behavior of the apoptosis cell in yeast. S. cerevisiae in capillary electrophoresis (CE), it was found that the differences between apoptosis cells and normal cells were showed obviously. The absorption peak at 300 nm for the apoptosis cells disappeared, and the absorption peak at 220 nm in 12 similar to 18 min for the apoptosis cell nucleus was increased. This experiment confirms that the dynamic detection and analysis for cell apoptosis process by. capillary electrophoresis are feasible.
