Chemopreventive effects of oltipraz on glandular stomach carcinogenesis in rats treated with N-methyl-N'-nitro-N-nitrosoguanidine

摘要
The effects of oltipraz on glandular stomach carcinogenesis induced by N-melhyl-N'-nitro-N-nitrosoguanidine (NBBG) were investigated in rats. A total of 120 male Wistar rats were divided into 6 groups. Rats in groups 1-3 were given 100ppm MNNG in their drinking water for 10 weeks and were respectively fed diets supplemented with 0.04%, 0.02% and 0% oltipraz for 12 weeks, starting at 1 week before the MNNG treatment. Groups 4-6 were similarly treated with oltipraz without the carcinogen exposure. At the 81 st week, ail surviving animals were autopsied. The incidence and multiplicity of adenocarcinomas were significantly (p<0.01) lower in group 1 than in group 3. The multiplicity of atypical hyperplasias in the pyloric region was also significantly (p<0.05) smaller in group 1 than in group 3. Our results thus indicate that oltipraz inhibits induction of proliferative glandular stomach lesions by MNNG in the rat.
