Production of transgenic hygromycin resistant carnation (Dianthus caryophyllus L.) plants after cocultivation with Agrobacterium tumefaciens

摘要
Agrobacterium-mediated transformation procedure was developed for petals and basal parts of carnation leaves using the hygromycin resistance gene (Hpt) as a selective agent. Formation frequency of hygromycin positive clones varied from 0,5% (petals and leaves of greenhouse plants) to 11,5% (leaves of in vitro plants). PCR analysis confirmed presence of Hpt gene in all recovered plants. Expression of GUS gene was detected in 95% lines raised from leaves of in vitro plant, in 67% lines raised from leaves of greenhouse plant and in 50% lines obtained from petals.
