In Vitro Chemotherapy of Pear Cultivars

摘要
The aim of the present study was to determine whether chemotherapy with ribavirin can be used to eliminate Apple stem pitting virus (ASPV) from in vitro plants of infected pear cultivars 'Astra', 'David' and 'Erika'. According to RT-PCR testing, selected field-grown trees of all pear cultivars were infected with ASPV. In vitro cultures were successfully multiplied on MS medium with BAP (6-benzylaminopurine). Ribavirin was used at 20 mg L-1 for virus elimination. Low phytotoxicity of ribavirin at 20 mg L-1 was observed only for the cultivar 'David'. Six of the twenty shoot tips of 'David' showed apex necrosis and died after transfer to regeneration medium. The success of chemotherapy after RT-PCR testing of ribavirin treated plants varied between 65.0% for cultivars 'Astra' and 'Erika' and 71.4% for cultivar 'David'. The results achieved and the relatively high percentage of virus free plants obtained after 6 months of in vitro cultivation confirm the effectiveness of ribavirin in eliminating ASPV in pear.
