GENETIC DIVERSITY OF LILIUM LANCIFOLIUM GERMPLASM RESOURCES REVEALED BY ISSR AND SRAP MARKERS

Lilium lancifolium is the only wild triploid species of genus Lilium with ornamental value, as well as edible and medicinal potential. China has the vast territory and changeable climate and it is an important distribution area for L. lancifolium germplasm resources. L.lancifolium tolerates a variety of habitats due to its high ecological adaptability. At present, not much is known about genetic diversity of L. lancifolium native to China. This present study first detected the genetic diversity of 74 L. lancifolium samples from different regions of China using ISSR and SRAP markers and then investigated the effect of eco-geographical factors on genetic diversity of tiger lily. Finally, 131 and 167 bands were amplified by ISSR and SRAP marker with 10 sets of primer of each marker, the level of genetic diversity revealed by ISSR (PPB = 49.62%, H = 0.1398, I = 0.2141) was slightly higher than SRAP marker (PPB = 47.90%, H = 0.1021, I = 0.1628). Cluster analysis for all the materials was carried out based on unweighted pair-group method with arithmetic means and principal coordinate analysis. Cluster results showed that all the materials were felled into three groups and the differences between groups were significant (p<0.0001). High degree of differentiation were also detected (Fst = 0.243/0.376). In ISSR marker, altitude, annual mean temperature and annual mean precipitation explained 16%, 20% and 19% of the total variance, respectively, while in SRAP marker, they explained 24%, 19% and 16% of the total variance, respectively. It was suggested that genetic variation of L. lancifolium was significantly influenced by eco-geographical factors. Meanwhile, the differences between the corresponding groups, grouping based on the eco-geographic factors, were significant too. Our results indicated that both ISSR and SRAP markers can effectively reveal the genetic diversity and differentiation of L. lancifolium germplasm. Considering the significant impact of eco-geographical factors, in situ conservation strategy should be taken. On the other hand, we need to conduct systematic collection, comparing and screening for L. lancifolium gerplasm in order to make a better use of it in the future.
