Effect of tumor necrosis factor-alpha (TNF-α) on erythropoietin induced erythroid cell formation

Tumor necrosis factor-alpha (TNF-alpha) was administered to exhypoxic polycythemic mice to investigate its effect upon erythropoietin (EPO) induced erythroid cell formation, as monitored by Fe-59 incorporation into circulating red blood cells. TNF-alpha injection suppressed erythroid cell formation induced by EPO in dose related manner and its administration 6 hour prior to EPO produced the maximum effect. This indicated that the effect of TNF-alpha was more on the cells which respond to EPO than on the cells produced by EPO. Higher doses of EPO did not abolish this suppression. These results confirm the inhibitory action of TNF-alpha upon erythroid cell formation which might lead to the anemia in the long term.
