Effects of solution behavior on polysaccharide structure and inhibitory of α-glucosidase activity from Cordyceps militaris

摘要
Crude polysaccharide of Cordyceps militaris was obtained by hot water extraction, Sevag deproteinization and alcohol precipitation, and then purified by Sephadex G-200 column chromatography. The extraction purified was named as CPS. The effects of pH, Ca2+ and DMSO on the structure of the CPS were analyzed by HPLC, Congo red assay, circular dichroism spectra and Atomic force microscopy (AFM) respectively. The results showed that the great changes of the retention time, spatial structure, molecular asymmetry and surface morphology of CPS have taken place. The inhibition rate of alpha-glucosidase of CPS treated by different solutions was reduced, which illustrated that the advanced structure of CPS affected its alpha-glucosidase inhibitory activities.
