Tyrosinase and TRP2 expression in hipomelanotic malignant melanoma

Amelanotic malignant melanoma (MM) can be sometimes difficult to diagnose both on H&E slides and immunohistochemical (1HC) tests. We analyzed the expression of S100 protein, HMB45, Melan A, tyrosine and TRP2 in 20 nodular pigmented MM, 4 hipo/amelanotic MM and 2 malignant peripheral nerve sheath tumor (MPNST). S 100 protein, HMB45 and melan A were unevenly distributed within the tumoral mass in MMs, with two hipomelanotic MMs with focal positivity for S 100 protein or very few positive cells for HMB45. Tyrosinase and TRP2 showed intense diffuse pattern of positivity in hipomelanotic MMs. None of the MPNSTs had positivity for either tyrosinase or TRP2. Tyrosinase and TRP2 are useful markers to be added to the IHC panel of antibodies of a hipo/amelanotic MM. Also, they may be used to exclude a MPNST.
