MIR-328-5P INHIBITS WNT/B-CATENIN TO PROMOTE THE PROGRESSION OF INTERVERTEBRAL DISC DEGENERATION

摘要
Introduction: Intervertebral disc degeneration (IDD) is a common orthopaedic disease, which has seriously thrown the health of gerontal patient. MiR-328-5p involves in the progression of multiple diseases, while its function and regulation mechanism on IDD remain unclear. Methods: In this study, the pathological tissues were used to observe the expressive difference of miR-328-5p. Moreover, qRT-PCR, western blot, CCK-8 assay, dual-luciferase reporter gene assay and flow cytometry assay were used to explore the effects of miR-328-5p on human intervertebral disc nucleus pulposus cells (HIDNPCs) and reveal the pathogenic mechanisms of miR-328-5p on IDD. Results: The results showed that miR-328-5p was significantly upregulated in IDD tissues, and increased miR-328-5p could remarkably suppress the viability and promote apoptosis of HIDNPCs. The study found that ACAN was a target of miR-328-5p, and ACAN upregulation could significantly reverse the effects of miR-328-5p on HIDNPCs. Besides, it was also found ACAN could activate the Wnt/beta-catenin in HIDNPCs while overexpressed miR-328-5p could reduce the activity of Wnt/beta-catenin via inhibiting the expression of ACAN. Conclusion: In conclusion, this study suggests that miR-328-5p inhibits Wnt/beta-catenin to promote the progression of IDD via targeting ACAN.
