Proteomic analysis of renal cell carcinoma and kidney control tissues

摘要
This study was to establish high resolution two-dimensional electrophoresis profiles for renal cell carcinoma (RCC) tissue and paired tumor-adjacent kidney tissue, and to identify the differential expression. proteins. The total proteins were isolated from histological study confirmed RCC and paired tumor-adjacent kidney tissues and were separated by two-dimensional gel electrophoresis (2-DE) followed by analysis using Image Master. 2D software. The proteins were identified by peptide mass fingerprint (PMF) and database searching. As a result, well-resolved and reproducible 2-D maps of analyzed tissues were obtained. Some of different proteins from RCC tissues were identified by mass spectrometry ( MS) when compared with paired tumor-adjacent kidney tissue, and these identified proteins were matched with protein profiles within SWISS-2DPAGE database. This study established and analyzed the expression maps of proteins, and demonstrated that the protein expression was significant different between RCC and tumor-adjacent kidney tissue. These data would be valuable for evaluation of underline mechanisms of tumor development and metastasis, and for establishment of diagnostic markers as well as targets for therapeutic intervention.
