TNF alpha and IL-10 gene expression in ischemic stroke

Significant role of pro- and anti-inflammatory cytokines in modulation of the immune response associated with ischemic stroke was documented. The source of cytokines in stroke could be ischemic brain cells as well as endothelial cells and peripheral blood leukocytes. The aim of this study was to evaluate the kinetics of TNF alpha and IL-10 gene expression in peripheral blood cells of the patients with ischemic stroke. By the method of semi quantitative RT-PCR with the use of plasmid as a competitor and GAPDH as a housekeeping gene we investigated TNF alpha and IL-10 mRNA levels in peripheral blood of 38 stroke patients. The temporal profile of investigated cytokines support the development of systemic inflammation resulting from stroke incidence and confirm the presence of negative feedback control between the production of TNF alpha and IL-10.
