Galectin-7 in the stratum corneum: a biomarker of the skin barrier function

摘要
ObjectiveSkin barrier disruption often occurs in diseased and damaged skin conditions such as atopic dermatitis (AD). We focused the galectin-7 protein (Gal-7) as a biomarker of skin condition and assessed whether the content of Gal-7 in stratum corneum (scGal-7) could be used as an indicator of skin barrier disruption and as an index of local skin symptoms in AD patients. MethodsAlteration of Gal-7 expression levels in keratinocyte and scGal-7 contents after barrier disruption by sodium dodecyl sulphate were evaluated in vitro and in vivo, respectively. Correlation between scGal-7 content and transepidermal water loss (TEWL) was examined in 126 healthy subjects. We performed single measurements of scGal-7 contents in 34 AD patients and serial measurements of 15 inpatients among them. SC samples were collected by the tape-stripping method, and scGal-7 content was determined using enzyme-linked immunosorbent assay. ResultsGal-7 expression in keratinocytes increased after barrier disruption. The scGal-7 content reflected the disruption of the skin barrier. The scGal-7 contents and TEWL values correlated in healthy subjects. The scGal-7 level was higher in AD patients than in healthy subjects. The scGal-7 contents in the cheek and neck of AD patients significantly correlated with the total and local skin lesion severity scores. Serial measurements in the inpatients showed that the scGal-7 contents in the cheek and neck decreased in tandem with local severity scores in response to treatment. ConclusionMeasurement of scGal-7 content in tape-stripped samples was useful for the evaluation of the skin barrier function in dry skin conditions such as AD. Resume ObjectifLa rupture de la barriere de la peau se produit souvent dans des conditions de la peau malade et endommagee telles que la dermatite atopique (AD). Nous nous sommes interesses a la proteine galectine-7 (Gal-7) en tant que biomarqueur de l'etat de la peau et avons evalue si le contenu de Gal-7 dans la couche cornee (scGal-7) pouvait etre utilise comme un indicateur de rupture de la barriere de la peau et comme un indice de symptomes cutanes locaux chez les patients atteints de la DA. MethodesL'alteration des niveaux d'expression de Gal-7 dans des keratinocytes et la teneur en scGal-7 apres rupture de la barriere par le dodecylsulfate de sodium ont ete evalues in vitro et in vivo, respectivement. La correlation entre la teneur en scGal-7 et la perte d'eau transepidermique (TEWL) a ete examinee dans 126 sujets sains. Nous avons effectue des mesures simples des teneurs en scGal-7 dans 34 patients DA et des mesures en serie sur 15 patients hospitalises parmi eux. Les echantillons du SC ont ete recueillis par la methode d'adhesifs arraches et la teneur en scGal-7 a ete determinee en utilisant un dosage immuno-enzymatique. ResultatsL'expression de Gal-7 dans les keratinocytes augmente apres rupture de la barriere. La teneur en scGal-7 reflete la perturbation de la barriere cutanee. Les teneurs en scGal-7 et les valeurs TEWL sont correlees chez les sujets sains. Le niveau de scGal-7 etait plus eleve chez les patients AD que chez les sujets sains. Les teneurs en scGal-7 dans la joue et le cou des patients atteints de DA sont significativement correles avec les scores totaux et locales de gravite des lesions de la peau. Des mesures en serie chez les patients hospitalises ont montre que les teneurs en scGal-7 dans la joue et le cou ont diminue en tandem avec des scores de gravite locales en reponse au traitement. ConclusionLa mesure de la teneur scGal-7 dans des echantillons d'adhesifs est utile pour l'evaluation de la fonction barriere de la peau dans des conditions de peau seche, tels que DA.
