Antiepileptic drugs and pregnancy: brain malformations in rats prenatally exposed to lamotrigine, vigabatrin and valproate

The treatment of epilepsy during pregnancy entails a number of concerns. While maternal seizures may adversely affect maternal and fetal outcome, prenatal exposure to antiepileptic drugs may increase the incidence of congenital abnormalities and affect postnatal cognitive development. In two recent studies, we have reported an increased incidence of cortical and hippocampal malformations in rats exposed to lamotrigine, vigabatrin and valproate during embryonic life. These results, as well as other experimental data, are presented here and their clinical relevance and implications are discussed.
